Delaware Route 16 is an east/west state highway in Delaware, mainly across northern Sussex County, with a small portion near the Maryland border in extreme southwestern Kent County. It runs from Maryland Route 16 at the Maryland border near Hickman to the Delaware Bay at Broadkill Beach. The state of Delaware has encouraged its use as an alternative route for Delaware Route 404 to avoid potentially costly upgrades to Route 404. Route 16 serves mostly to connect the Baltimore-Washington Metropolitan Area with the ocean resorts via U.S. Route 50 in Maryland and Maryland Route 404.

Upon the road's start at the Maryland line in Kent County, the road is named Hickman Rd after the small community of Hickman that straddles the Maryland and Delaware lines. After connecting with Delaware Route 36 west of Greenwood, it adopts that roads name of Scotts Store Rd. for approximately one mile until it enters the corporate limits of Greenwood. At this time it adopts the town street name of Market Street which it continues until leaving the corporate limits. The route intersects U.S. Route 13 in the eastern part of Greenwood. At the eastern corporate limits, Delaware Route 36 breaks off heading northeast towards Milford while Route 16 continues east towards Ellendale and the resort areas. Known originally as the Ellendale - Greenwood Highway, DelDOT has since changed the name of Route 16 between Greenwood and Ellendale to Beach Highway to reflect the desire of DelDOT for travelers to use this as an alternate route to the beaches and relieve congestion on Delaware 404. Upon its intersection with U.S. Route 113 in Ellendale the resort area of Delaware begins. This intersection is considered the northwest corner and Gateway to Delaware's Resort Beaches. In the corporate limits of Ellendale, Route 16 becomes Main Street and upon leaving becomes Milton - Ellendale Highway. Upon crossing Delaware Route 5 (Union Street) in Milton, the name changes for the final time to Broadkill Road. The route intersects Delaware Route 1 and continues east until it terminates at the Delaware Bay.
